Cover: X-Gal staining of brains derived from transgenic mice harboring a Tie2 promoter-tetracycline-on cassette, and a nuclear localized form of -galactosidase under control of a tetracycline-responsive promoter. This system allows for regulated, endothelial-selective expression of the nl-Gal transgene in response to exogenous doxycycline. The brain on the right was derived from an animal treated with doxycycline, and the brain on the left was from an untreated animal. The X-Gal staining is seen in the nuclei of the endothelial cells lining the cerebral vessels on the ventral surface of the brain, in the doxycycline-treated animal. This system can be readily adapted to selectively express any transgene of interest in the vascular endothelium.
